Bulk Discount: To meet your high throughput needs, we offer significant discounts on bulk orders (> 10 service items)
for research labs in pharmaceutical and biotech companies and research institutes. All bulk orders will be delivered within
designated time whether in a single P.O. or over a period of time.
Outsourcing and Collaboration: Our flexible custom services are also provided to research labs on a contract basis. In
a contract service, LBL scientists will offer you free consultation along with our high quality service products. We welcome both

RNAi Target Selection
Target selection is critical for gene silencing by RNAi technology and currently a trial-and-error process. The
RNAlysis™ RNAi Target Selection Protocol developed by LBL scientists involves
computerized automated screening of potential target sites according to our most recently generated thermodynamic-based selection
algorithm and semi-automatic multiple-alignment procedure to minimize off-target effect.
Using this Protocol, we are confident to provide you two most effective RNAi target sites on a single target gene. If result
from one of the two target sites is not satisfied, we will provide you the third one without additional charge.
If you have special requirements for your RNAi assay similar to but not limited to the following conditions, let us know. We
will meet your needs without additional charge:
- The target gene has sequence-related family members, and you want to have a specific knockdown on the target gene without
affecting other family members; Or
- You want to knockdown all or some selective genes in a gene family.
- The target gene has isoforms, and you want to have a specific knockdown on the selected isoform without affecting others; Or
- You want to knockdown all isoforms of a target gene.
The turn around time of this service is typically 2 to 3 days. Time may vary depending on the volume of order and complexity of
your gene.
This service is free for customers, who purchase additional services and/or products from LBL.

DNA Hairpin Cloning
Initial cloning of hairpin structure from synthetic DNA oligos is often problematic. LBL scientists have adapted technical
skills, by which we are able to provide reliable, cost-effective DNA hairpin cloning service for customers who are using
RNAi Combination Toolbox™.
In this service, we synthesize the DNA hairpin and clone it into one of our vectors of your choice.
The construct will be verified by DNA sequencing. The final plasmid product (1 - 10 µg) will be sent to you together with
the sequence report.
Specify the name and catalog number of the vector when you make a quotation. If you have already purchased the vector or you
have used this service on the same vector before, provide proof of purchase (e.g. the reference number on previous invoice)
to avoid repeated charge on the vector that will be used.
If you are using our RNAi Target Selection service combined with this service and result from one
of the two target sites is not satisfied, we will make the third one for you without additional charge.
If you have a validated target sequence that you want to subclone into one of our vectors, we will do the cloning for you
without additional charge, given the conditions that:
- You provide the target sequence only, which is equal to or less than 29 nt; Or,
- You provide the sequence of DNA hairpin, of which the total length including sense, anti-sense and loop, is
equal to or less than 70 nt.

DNA Cloning
We can clone various DNA fragments into a plasmid vector provided or authorized by you. Simply provide your starting materials,
let us do the rest. Our DNA cloning services include but not limited to:
- Standard subcloning of a DNA insert fragment from one vector to another;
- N-terminal and C-terminal tag integration;
- Promoter-reporter cloning;
- Multiple cloning site changes;
Plasmid vector and DNA insert fragment will be prepared by restriction digestion or PCR with low cycle, high-fidelity
amplification from a template. After ligation and transformation, clones will be screened for the correct restriction fragments
and for orientation, if necessary. When PCR has been applied, product will be sequenced to ensure sequence integrity. Finally, you
will receive a mini-scale plasmid DNA preparation and a detailed project report.
Custom promoter: In LBL, we offer promoter subcloning service. These promoters,
which are derived from human, mouse and rat species, give you cutting-edge technology to study gene function in a tissue-specific
manner. Simply send us your gene of interest, we will swap the promoters for you.
Large insert/vector: We have expertise to perform long (> 4 kb) DNA fragment ligation and DNA subcloning with large size
(> 10 kb) vectors. By using our services, you will never become frustrated with difficult inserts and vectors anymore. Focus on your
research and move on.
Targeting vector construction: We also have expertise to prepare and handle extra large vectors, which are typically
30 - 50 kb in total, for gene targeting applications. We provide strategy design, isolation and characterization of genomic clone,
mapping and construction services of the targeting vector for your gene knock-in or knock-out studies. Please note, this service
is on contract-basis only.

De Novo PCR Cloning
If you are planning to work on a gene that you don't have, or if you want to have a genomic DNA fragment for study, we can
clone a gene or gene fragment for you from genomic/cDNA libraries of human, mouse and rat species. Genes from other species may also
be available on request.
Tell us the name or Genbank accession number of the gene that you are interested. We will design and supply the PCR
primers to amplify the gene, and subcloned it into the plasmid vector of your choice.
All PCR products will be sequenced (both DNA strands) to ensure sequence integrity. Finally, you will receive a mini-scale
plasmid DNA preparation and a detailed project report.
RT-PCR cloning: If you have a gene, a special EST or gene isoform that you know about the sequence, we will design and
supply the PCR primers to amplify the target sequence from RNA templates. Simply provide us
with DNA sequence information and tissue, cells or RNA samples, we will do the rest for you.

Site-directed Mutagenesis
We can carry out site-directed mutagenesis on the selected sequences to make specified base changes including deletions,
insertions and substitutions, into a gene of your interest.
All mutagenesis clones are sequenced to ensure that the desired base changes have been made and to ensure sequence integrity
of the remainder of the clone. Simply send us your clone and a list of the mutations you want. You will receive a mini-scale
plasmid DNA preparation and a detailed project report.

DNA Preparation
To provide a one-stop-shop service for your research project, we can carry out large-scale plasmid DNA preparation at standard
research grade. Our final products will be suitable for various applications including restriction digestion, DNA sequencing,
yeast and bacterial transformation, mammalian cell transfection, in vitro transcription and translation, etc.
Preparation starts from 500-µg scale. Larger scales or bulk preparations are available with reduced price.
The turn around time of this service is typically 5 - 7 days. Time may vary depending on the volume of order and customers

Cell Line Generation
In LBL, we have state-of-art mammalian cell culture facilities to provide you custom cell line generation services. Simply
send us your construct ready for transfection. You will receive three cloned cell lines or a polyclonal cell line of your choice.
Stable cell lines with gene silencing can also be made from RNAi Combination Toolbox vectors,
which are carrying selection markers. Expression of your gene or gene knockdown is guaranteed and will be verified by
immunoblotting.
Check out the list of commonly used cell lines. If your cells are not in the list,
send us your cells and we will do the rest for you.
If your gene has been conjugated with one of the following epitop tags, we will verify the expression of your gene by immunoblotting
without additional charges:

Yeast-Two Hybrid
In accordance with your molecular and cell biology study, we offer protein-protein interaction analysis using our advanced yeast-two hybrid
technologies:
Single Protein Interaction
If you have cDNAs that are already in yeast-two hybrid vectors, however encounting difficulty to optimize the cell growth
conditions, simply let us do the test for you. We will provide you the result with appropriate controls.
The turn around time of this service is typically 1 - 2 weeks. Time may vary depending on the volume of order and customers
will be informed in the Purchase Order Confirmation.
Library Screening
In LBL, we will screen one of our libraries or a library provided by a customer to identify proteins that
interact with a protein of interest. After the screen, you will receive minipreps of up to 20 interacting clones and clone analysis
including insert size, sequence and BLAST search results.
Screening steps are but not limited to:
- Bait self-activation test;
- Initial library screening;
- Validation and test of specificity;
- Sequencing of up to 20 interacting clones.
